And though it is not specifically mentioned what the Truthsayer Drug is, it is safe to assume that it is a form of Spice essence which is what they use in the Reverend Mother Tests correct? The Spice Agony as it were?

if it isn't that, it must be some other drug and not the Water of life because you are correct Ghanima, the Water of Life wasn't known until after Muad'ib.

Also, Sappho juice is utilized by mentats to give them clear thinking in their higher order calculations, so obviously it has some sort of mental enhancement properties. In the prequels Piter DeVries utilizes it to get visions of the future too, so it must be useful in some respects too.
So it is possible that there are other drugs out there that mimic the mental enhancements that the Spice can give.
