According to Leto 2, when Hwi asks him what HE believes, he says something like "I don't believe that nature creates something from nothing," or something along those lines. I took that to mean that leto believes that God actually did create the worms and mankind, and that he really is doing God's work.
There is other life in the universee, but not likely other "intelligent" life. That, according to the books. Herbert mentioned that there is other life on some planets, but it certainly does appear that earth has colonized most of the planets with Terran animals and plants or some sort of mutated form of them.
There is also the fear that one day humanity might finally encounter an alien intelligence that could be a threat to them. This is one of the reasons given for the Great Houses to maintain their stockpiles of nukes.
When you consider that the Empire spans MANY MANY galaxies, it's pretty over-whelming. Then the Scattering expands the empire a thousandfold(unlimited expansion, it actually says.)
