I believe the Reverend Mother test is described in the later books of Heretics and Chapterhouse and that the test includes a concentrated amount of Spice that is basically like poison. I'd have to reread to be sure, and I know others on this site know it better than me, but it is similar to the Water of Life transmutation. Yes it involves Spice.
