I always thought that was one of Joss' sideways and well-protracted jokes. Giles telling Buffy he doesn't have an instruction manual (which he really doesn't---not for a slayer who tries out for cheerleader and wants to date), then later casually talking about the slayer handbook with Kendra. Because you left out the line that connects the two:

Giles: After meeting you, Buffy, I realized that, uh, the handbook would be of no use in your case.
