Further investigation resolved the problem by creating a
Transform2 := TImageFormatTransform.Create and PNG2 := TPNGGraphic.Create and applying the transformation, then freeing them and then performing the code already posted. However, this is did not work in the main application so the problem still exists.

I found it interesting that between the If test and the loop in the GetIndex method the PNG.Palette.palNumEntries property becomes inaccessible.

I'm not sure about the praticality of changing from TBitmap TBitmapGraphic classes.
